The midfielder was linked with a January switch to Old Trafford, having impressed David Moyes with a string of outstanding performances in La Liga.
Koke has been a crucial factor in Atletico’s surprise title challenge, and United have been linked with a summer move despite the Spaniard having a contract until 2018.
But the 22-year-old has ruled out the possibility of a switch to the Premier League, insisting he wants to remain at Atletico and play under Diego Simeone for the long term.
“I have a long contract and I am not thinking about leaving,” Koke told Minuto 116.
“Atletico is my home. I am happy that Simeone has confidence in me and I want to keep working for him for many years.”
Moyes has been linked with a host of La Liga midfielders since taking over at Old Trafford last summer.
Thiago Alcantara, Cesc Fabregas and Anders Herrara all rejected the chance to move to United in the summer, while Koke appears to be the latest to snub their advances.
